Introduction
A mini preparation, also called a plasmid preparation, is a process through which plasmid DNA is extracted and purified. It is used to analyze bacterial clones for molecular cloning. Mini preparation first grows the bacteria culture through transformation and inoculation. During this step, rich medium (LB), such as LA or LK(ampicillin or kanamycin), is used. Then it cultivates and induces lysis of the bacteria. At last, it purifies the plasmid DNA that is extracted. Depending on the plasmid copy number, type and size, the bacterial strain, the growth conditions, and the kit, the plasmid DNA should go through the miniprep, midiprep, maxiprep, megaprep, and gigaprep.
Alkaline lysis method is the most commonly used. First, the selected bacterial colonies are grown in LB broth that has the selection antibiotic.
